GAS cysteine protease SpeB virulence factor cleaves GSDMA after Gln246 or Gsdma1 after Gln247, which could be abrogated by the cysteine protease inhibitor E64. The N-terminal domain of GSDMA/Gsdma1 (N-GSDMA/N-Gsdma1) could oligomerize to form pores in the cell membrane, further causing lysis and the release of proinflammatory factors, ultimately leading to local inflammation. In addition, SpeB also processes the IL-1β zymogen (pro-IL-1β) to mature IL-1β, which is released extracellular from the pores formed by N-GSDMA/N-Gsdma1.
