血管瞬时受体电位通道的生理功能及其异常导致的相关疾病

瞬时受体电位(transient receptor potential, TRP)通道最早由Cosens和Manning在果蝇突变体的光感受器细胞中发现。在哺乳动物中, TRP通道广泛表达于中枢和外周神经系统。绝大多数TRP离子通道为非电压依赖性、非选择性阳离子通道。在已经被鉴定的30余种存在于哺乳动物中的TRP通道亚型中, 有超过19种在血管中表达, 而血管中的TRP被认为可以通过调控Ca2+内流与释放, 参与调节多种生理功能, 包括调节血管张力、血管通透性、血管分泌功能, 参与机械信号传导等。某些TRP亚型(如TRPC3、TRPC6和TRPV1等)的异常表达与高血压或其他血管疾病密切相关。本文旨在对血管上的TRP通道的生理功能及其异常导致的相关疾病进行概述。

Transient receptor potential (TRP) genes were first described in the fruit fly Drosophila melanogaster.The TRP channels are widely present in central and peripheral nervous systems in mammalian.Most of the TRP channels are voltage independent and non selective cation channels.Among the 33 identified TRP channel proteins,at least 19 of them are expressed in blood vessel (mainly expressed in the vascular endothelial cells and vascular smooth muscle cells).These channels are responsible for the influx or efflux of Ca2+ and therefore contribute to vascular functions including regulation of vascular permeability,controlling vascular tone,mechanosensation and vascular secretion.Abnormal expression and/or dysfunction of TRP channels were observed in the hypertensive blood vessels and other vascular diseases.This article reviews the physiological function of the TRP channels in blood vessel and briefly describes the role of the TRP channels in developing vascular related diseases.

内皮型一氧化氮合酶翻译后调节的研究进展

内皮型一氧化氮合酶(endothelial nitric oxide synthase, eNOS)翻译后调节机制复杂, 可直接通过磷酸化、乙酰化等多种形式的修饰改变活性, 同时钙调蛋白、小凹蛋白、热休克蛋白90、G蛋白偶联受体、激酶、骨架蛋白等多种蛋白质可通过与eNOS直接或间接作用, 调节eNOS的活性;BH4和吡哆醇可通过影响脱偶联调控eNOS的活性。对eNOS调节机制的探讨将有利于发现防治内皮功能障碍相关疾病的新靶点。

The post translational regulatory mechanism of endothelial nitric oxide synthase (eNOS) is a complicated process concerning the regulation of its activity with multiple modifications,including phosphorylation,acetylation etc.Various proteins,such as calmodulin,caveolin,heat shock protein 90,G protein coupled receptors,kinases and cytoskeletal proteins showed direct or indirect interactions with eNOS to regulate its activity.In addition,BH4 and pyridoxine can also regulate the eNOS activity through eNOS uncoupling pathway.Studies about regulatory mechanism of eNOS would help us to find novel targets in prevention and treatment of endothelial dysfunctions related to various diseases.

转化医学作为基础与临床之间的桥梁发挥着非常重要的作用, 它的提出促进了基础研究与临床应用之间的互相转化。以络病理论创新指导临床重大疾病防治研究, 提高了临床诊疗水平, 带动系列国家专利新药研发, 络病理论教学建设与学术推广, 促进了创新成果转化和产业化发展, 从而又进一步推动了临床应用, 形成了“理论-临床-科研-教学-产业”五位一体的良性运行机制, 推动中医药产业的不断发展, 以及中医络病理论的继承与创新, 真正实现了理论、基础与临床的相互转化。

Translational medicine (TM) is a bridge to connect basic sciences with medical applications.The emergence of the concept of TM facilitated the transformation between basic scientific research and clinical application.In this article,we will discuss the roles of collateral disease theory in prevention and treatment of major diseases,improving the quality of the diagnosis and treatment,performing research and development (R&D) of new patent drugs,theoretical teaching and academic advancement,development of Chinese medical industry and clinical application,with the optimal operation of the “five in one” mode:‘basic theory—clinical application—scientific research—education—industrialization’.This process can in turn enhance the sustainable development of Chinese medical industry,promote the inheritance and innovation of collateral disease theory and implement the translation between medical theory,basic research and clinical application.
